ATG16L1 is a critical protein involved in autophagosome initiation. It binds to ATG12-ATG5 complex with E3-like ubiquitin ligase activity which is essential for LC3 conjugation with PE lipid (PMID: 23202584). With the help of WIPI2, the complex is localized on the membrane performing its critical function as conjugation machinery. Also, the N-terminal amphipathic helix is known to bind to the membrane (PMID: 30778222) facilitating ATG12-ATG5 complex tether LC3 proteins on phagophore though conjugation. Several reports suggest the colocalization of ULK1 complex and ATG12 conjugation system on the growing autophagosome via FIP200 (RB1CC1) (PMID: 23392225, 23262492).
