ATG16L2 has structural as well as functional similarities with ATG16L1 which is key component of LC3 conjugation machinery. The conjugation machinery plays a critical role in covalently attaching PE lipids to the LC3 proteins during the initial stages of autophagy. ATG16L2 can exist as either a homodimer or a heterodimer (in complex with ATG16L1). It has been shown that the lack of ATG16L2 in mouse embryonic fibroblasts does not impair the progress of the autophagic pathway (PMID: 22082872).
