ATG2B belongs to ATG2 family of proteins having a characteristic property of tethering the edge of isolation membrane (IM) to ER exit site (ERES) and mediating lipid transfer (LT) from ERES to IM for IM expansion at the same time (PMID: 30911189). This activity is crucial to autophagosome biogenesis. ATG2B possesses both the lipid transfer and membrane tethering activity which are promoted by negatively charged membranes and WIPI4. Studies suggest that the LT activity is first enhanced by PI3P itself and is further enhanced by WIPI4. Moreover, the LT activity is conserved among species although the regulation mechanism might have diversified during evolution (PMID: 31721365).
