Phophatidylinositol-3-kinase class-III belongs to the family of lipid kinases that are involved in membrane trafficking. PIK3C3 is recruited with other elements of autophagy forming PIK3C3 complex during autophagy initiation. Along with PIK3C3, the complex comprises Beclin-1, UVRAG, and ATG14. The kinase activity of PIK3C3 is responsible for phosphorylating phosphatidylinositol to phosphatidylinositol-3 phosphate (PI3P) (PMID: 7628435). This phosphorylation helps autophagic proteins such as WIPI recognize the specific lipids and promotes further events in autophagy (PMID: 21548784).
