WIPI4 belongs to the PROPPIN family of proteins with beta-propeller fold essential for binding to phosphoinositide (PtdIns3P) lipids. While WIPI1/2 are well known to bind to autophagosomal membrane and eventually recruit the conjugation machinery (ATG12-ATG5-ATG16L1 complex) (PMID: 24954904), WIPI4 associates with ATG2 (a lipid transferring protein), and this complex has the ability to specifically mediate membrane tethering which is essential for autophagosome biognesis (PMID: 30275332)
